Heroin addicts develop tolerance of teh drug after sometimes because when the drug binds to opiods receptors it triggers the enzymes which fire the impulses after sometime the repeated activation of the opiod receptor the enzymes adapts to the drug and stops responding and also stops firing the impuses and it causes reduced reaction to the drug.

So they increase the dosage of drug to get the reaction thus addicted person can tolerate the drug quantity which can kill other person who does not regularly use the drug.
