Introduction to enzyme kinetics: -

Enzyme kinetics is always studied taking any one enzyme as base. In the given experiment, we are taking the enzyme phosphatase as base. Phosphate reacts with colorless p-nitrophenol phosphate (NPP) substrate, to give yellow p-nitrophenol as product. Now, if we have to find out the optimum enzyme concentration and optimum substrate concentration, then we will have to vary the enzyme and substrate concentration in two different experiments.

In the experiment where we are finding the optimum substrate concentration; we will take 8 test tubes with varying concentration of substrate, but with the same concentration of enzyme. As blank we will keep the enzyme solution, but will not add the substrate.

On the other hand, in the experiment where we are finding the optimum enzyme concentration; we will take 8 test tubes with varying concentration of enzyme, but with the same concentration of substrate. In the test tube taken as blank, we will add the substrate but will not add the enzyme.

In all cases, O.D. will be noted down using UV-Vis spectrophotometer at 575nm.
