8) A because,Zinc-finger nucleases (ZFNs) are enzymes got by fusing a zinc finger to a DNA cleavage enzyme called a Fokl.

B BECAUSE ,Engineered zinc finger nucleases (ZFNs) induce DNA double-strand breaks at specific recognition sequences and can promote efficient introduction of desired insertions, deletions or substitutions at or near the cut site via homology-directed repair (HDR) with a double- and/or single-stranded donor DNA template. However, mutagenic events caused by error-prone non-homologous end-joining (NHEJ)-mediated repair are introduced with equal or higher frequency at the nuclease cleavage site. Furthermore, unintended mutations can also result from NHEJ-mediated repair of off-target nuclease cleavage sites.

9)A BECAUSE TALENs are produced by fusing the TAL DNA effector (TALE) binding domain to a Fokl catalytic domain DNA cleavage domain.

B because this leads to mutagenesis and target gene replacement

C because TALENs technology can, in theory, edit any portion of the genome.

10)B because When making modifications to a gene using CRISPR, the population of transfected cells will contain a combination of NHEJ-repaired and HDR-repaired alleles.

C because CRISPR-Cas9 can target DNA sequences with only a 70% probability of success given a random stretch of DNA sequence

D because  CRISPR In Prokaryotes This defense mechanism of prokaryotes is used as editing technique which can used in gene therapy process as well. The CRISPR Cas editing method makes the use gRNA for identification and cleavage of DNA strands.

    4. The CRISPR-Cas9 system is an important new technique in molecular biology. What is the natural function of this system? Describe how you would use this system to generate a null mutation in another organism (i.e. explain Figure 6-43). How does it work? What is the modification of the method that allows for correction of a mutation (e.g. the mouse crystalline gene)? And lastly, what are the problems with the CRISPR system?
