Answer #1

The plasmid is cut by EcoRI to obtain 14.5kb fragment. HindIII cuts the plasmid to produce 15.5kb fragment. if both EcoRI and HindIII cut the plasmid 1Kb fragment is to be obtained. There is no possibility to obtain 0.5Kb fragment. Therefore, option A is correct.

please help me with these questions Lab 8 Extension Activity: Plasmid Mapping and Restriction Enzymes Mapping the Plasmid The first step in mapping a plasmid is to determine how many times a restriction site is found on that plasmid. Examine the results for plasmid 55 as an example. The data given in the following table are for the double digest using EcoRI and Pstl. Also, giving are the data for single digests by the individual enzymes. The numbers in the...

You have accidentally torn the labels off two tubes, each containing a different plasmid, and now do not know which plasmid is in which tube. Fortunately, you have restriction maps for both plasmids, shown in the figure below. You have the opportunity to test just one sample from one of your tubes. You have equipment for agarose-gel electrophoresis, a standard set of DNA size markers, and the necessary restriction enzymes. A)Why won’t making a single cut (e.g. with HindIII) help...

Can someone please help me answer these? Neat work is greatly appreciated. Explanation will also be appreciated (optional, if you have time). Thank you so much! I will give a big thumbs up :) (1) You start with the pUC18 plasmid that is a -2.7 kb circular DNA molecule. The multiple cloning site (i.e., polylynker) has unique restriction sites in the following order: HindIII, BamHI, EcoRI. Recall that unique restriction site means that these sites are only found once on...
