Rb acts as a tumor suppressor gene. The presence of un-phosphorylated active form of Rb inhibits cell cycle by preventing DNA replication. When a normal cell requires undergo cell division, Rb (as a substrate) is phosphorylated by cdk2 and thus inactivated. Inactivation of Rb through phosphorylation triggers the cells to enter cell cycle.
