There are a few strategies to remove the GFP from PF moro Green vector: one of them is by Restriction enzymes.

As you can see

On both sides of GFP gene 2 Restriction enzymes are present . They are $\rightarrow$

1) Bam H1 (2) xba 1.

These 2 R.E are mixed with this vector,

• Graenally the GFP gene portion is cut off from the plasmid.
• Another gene containing both the R,E eites also mixed with the vector.
• Then, the other gene gets attached in the site of GFP , due to the same Restriction sites presence.

Thus, GFP is removed so that it could be directionally cloned into a new cector.
