E + S ↔ [ES] →    E + P      (enzyme catalytic reaction)

Where,

E = Enzyme, S = substrate, P = product.

Michaelis-Menten equation: Vo = Vmax [s]/ km + [S]

where,

Vmax = maximum velocity of the reaction, Vo = velocity of the reaction, Km = michaelis constant, [S] = substrate concentration.

Now let us suppose the Km = substrate concentration

Then, Vo = Vmax [S] / 2[S]

Or, V0 = Vmax/2

Vo 7 Vmax/2 [S]

Kcat is the turnover number of an enzyme.

Kcat = Vmax/ [E]T

[E]T   = concentration of an enzyme

Km/Kcat = catalytic efficiency.
