Predicted Phenotype- Non- Tester.

Will Your PCR product be Digested with HaeIII enzyme ? - No. Bacuse this Restriction site is only prasent in the tester allele of the TAS2R38 gene.

Based on the Hypothesis above do you expect a band around 203bp for your digested Sample- Yes. If i am a true Non-tester i will have a band at 203bp.

Do You expect a band on 156bp- No. bacsue me being a non tester, my DNA will lack the restriction site at TAS2R38 locus. so there will be no cleavage by HaeIII enzyme and there will be no band at 156 bp (while for True testers and moderate testers there will be a HaeIII cut site resulting in the presence of 156bp band in the DNA electrophoreisis gel).

Case 3- If my wife is a tester (TT)

All the children will be heterozygous (Tt).
