The transport system that conveys malate and alpha-ketoglutarate across the inner mitochondrial membrane is inhibited by n-butylmalonate. Suppose n-butylmalonate is added to an aerobic suspension of kidney cells using glucose exclusively as fuel. Predict the effect of this inhibitor on:

Answer #1

Ans:

(a) When n -butylmalonate an inhibitor of the malate-oaa shuttle, is added to kidney cells in presence of glucose, NADH accumulates in the cytosol as a result glycolysis operate anaerobically.

(b) Due to the inhibitor an increased rate of fructose diphosphate and fructose-6-P recycling, resulting in a wasteful use of ATP, for the maintenance of high oxygen consumption rates despite decreased net glucose formation.

(c) Lactate formation increase.

(d) as NADH accumulates in cytosol cannot get in the mitochodria for ETC so ATP synthesis is also decrease.
