Answer #1

From the first cell extract we infer that both protein A and B are membrane proteins since they are present in the menbrane of the normal cells.

Also,

Membrane proteins are of two types peripheral and intergral proteins.

Peripheral proteins are present on the inner or outer surface of the membrane and integral proteins are present buried deep inside the membrane.Peripheral proteins asy come out dissociate during the process of centrifugation whereas integral proteins do not and stay within the membrane .

And after treatment with urea if is found that protein B is present in the supernatant .

So from the factors mentioned and the observation Protein A can be identified as an integral membrane protein and Protein B as a peripheral membrane protein.

As in the mutants simce these proteins are altered results in the altered membrane integrity and thereby has lead to their shape change once again proving that A and B are membrane proteins and has a definite role in the maintenance of the shape of the cell.

6a. You have found a new protein of interest: this protein has a transmembrane domain (as predicted by hydropathy analysis). You want to confirm this and determine which portion of the protein is outside the cell and which portion is inside the cell. You begin with a protease protection assay, the results are below. Does this protein most likely have a transmembrane domain? Is the larger portion inside the cell or outside the cell? Is there anything wrong with the...

please help me!!! 5.) You have discovered mutants in two new gene mutants in Drosophila. One mutant, curl, has small, curly, nonfunctional wings. The other mutant, big, has enormous eyes. You decide to test if these two new genes are on chromosome #2, so you perform a three-gene linkage testcross using a gene that you know is on chromosome #2, trp, a mutant that cannot make its own tryptophan (and so must get it from its food). In all three...

please answer all that you can 1. You have genetically engineered green fluorescent protein (GFP) containing a KDEL sequence (GFP-KDEL). When GFP-KDEL is expressed in normal human fibroblasts and examined using fluorescence microscopy, the fluorescence appears diffuse across the cytoplasm. How would you explain this observations given that KDEL is supposed to be an ER-specific sorting sequence? A. This engineered GFP would not have a hydrophobic signal sequence to get it into the RER in the first place. B. The...

    2. A dominant allele H reduces the number of body bristles that Drosophila flies have, giving rise to a “hairless” phenotype. In the homozygous condition, H is lethal. An independently assorting dominant allele S has no effect on bristle number except in the presence of H, in which case a single dose of S suppresses the hairless phenotype, thus restoring the "hairy" phenotype. However, S also is lethal in the homozygous (S/S) condition. What ratio of hairy to hairless flies...
