2) There are numerous targeted therapies aimed at each of the hallmarks, but the soberly reality is that most target therapies do not work very long. For example, Epidermal Growth Factor Receptor (EGFR) inhibitors are effective for a while. Why is that so? Guide: Consider what could be responses from the tumor cells as well as the effect of neighboring cells.

Answer #1

Ans. The major reason rendering the target therapy inefficient is the continous mutation of the target cell. Owing to which the targetted therapy would not be able to ellicit the similar effect which it use to impart previously. In case of tumor cells the change in signalling system has been a major contributor to such response.

In this course, we, from time to time, talk about the use of so-called “targeted” anticancer therapies. These therapies are designed to target some specific aspect of the biology of the tumor that is different vs. normal, non-transformed cells. Such therapies, like farnesyltransferase inhibitors, Herceptin, etc. should theoretically speaking, provide the advantage of selectively neutralizing/killing tumors without adversely affecting non-cancerous cells in the body. This would, ideally, result in the elimination of cancer (i.e., effect a “cure”) with very few...

    2. A dominant allele H reduces the number of body bristles that Drosophila flies have, giving rise to a “hairless” phenotype. In the homozygous condition, H is lethal. An independently assorting dominant allele S has no effect on bristle number except in the presence of H, in which case a single dose of S suppresses the hairless phenotype, thus restoring the "hairy" phenotype. However, S also is lethal in the homozygous (S/S) condition. What ratio of hairy to hairless flies...
