The cell membrane of live cells is a dynamic structure and they are always moving at each and every time point. But if the environmental temperature is much lower than the ambient, then the dynamicity of the membrane loses. This is what happening in the above-said case. When the temperature condition is 4 degree then the membrane is not that dynamic and there is an even distribution of receptors observed by antibody staining throughout the cell surface. But when the temperature is 22 degree the receptors are localized in a specific place and when the receptors are aggregated at a specific area, many antibodies will bind at that region. Due to the bivalent nature of antibodies, the staining seems much higher than that of the antigen present. This is what happened in 22 degree incubation case.
