Answer: DNA molecule contain regions of nucleotide which is known as promoter that direct the RNA polymerse to begin the transcription . In prokaryotic cell two type of consensus sequences .These sequences are

1: TATAAT: This sequence is known as pribnow box. This box consist of consensus sequences (sequence which do not change) that is TATAAT which is present 10 nucleotide upstream of bacterial(prokaryotic) gene. As this sequence is present 10 units(nucleotide) left to bacterial gene so we put minus sign(as -10 upstream)

2: TTGACA; This is another consensus sequences present in prokaryotic gene. This is found 35 nucleotide upstream to bacterial gene

The sigma(σ ) factor present in prokaryotic RNA polymerase can detect this specific  consensus sequence either -10 or -35 upstream so as to initiate the process of transcription.
