the Michaelis-Menton equation is

V0= Vmax[S]/([S]+Km)

the Line-Weaver Burk equation is the reciprocal of the Michaelis-Menton equation

that is

1/Vo= Km/Vmax[S] + [S]/Vmax[S]

1/Vo=Km/Vmax[S] + 1/Vmax

this is in the format

y=mx+c

m is the slope of the graph that is m= Km/Vmax

c is the y intercept c= 1/Vmax

the x intercept of the graph is -1/Km

so using Line-Weaver Burk plot we can determine the Km and Vmax using X intercept, y intercept and slope of the graph so using Line-Weaver Burk plot is easier to calculate Km and Vmax.
