Need the answers to all please :)

1) You recently purchased a new mitogen at 1mg/1ml (stock) and then prepared 1:100 dilutions as a working solution (tube A). The company also suggest verifying the optimal dilution for your experiment by doing serial (twofold) dilutions.

Describe how you would prepare 500microliters of working solution (tube A).

What volume of inhibitor will you add in microliter?

You would like to optimize the working concentration of the inhibitor, and therefore perform two-fold dilutions of the inhibitor stock solution, tube A (1:100) and test those in your experiment. Prepare a set of 4 tubes for two-fold serial dilutions, each with a final volume of 100 microliters

In order to make 100 microliter of tube B, take 100 microliter of solution from tube A and dilute till 200 microliter.

In order to make 100 microliter of tube C, take 100 microliter of solution from tube B and dilute till 200 microliter.

In order to make 100 microliter of tube D, take 100 microliter of solution from tube C and dilute till 200 microliter.

In order to make 100 microliter of tube E, take 100 microliter of solution from tube D and dilute till 200 microliter.

By using this method you end up with 100 microliter of tube B,C and D but using 100 microliter out of the 200 microliter. made to make the next dilution. However, you still end up with 200 microliter of tube E instead of the recommended 100 microliter. If you did it with 50 microliter of the solution and diluted till 100 microliter, you would end up with 50 microliter of tube B,C and D but you would have 100 microliter of tube E.

3) The final dilution of tube C is 1:400

4) The working solution (tube A) has 10 microgram/ml

Due to two-fold dilution the tube B has half the concentration of tube A i.e. 5 microgram/ml

Similarly tube C has half the concentration of tube B i.e. 2.5 microgram/ml
