Amylase is an enzyme which is not used in molecular amplification methods.

DNA Ligase is widely used in many molecular amplification methods like ligase chain reaction and others, to join two or more nucleotides with one another which can be amplified later.

Reverse transcriptase is one of the most important enzymes used in molecular amplification methods, importantly RT-PCR. This technique is widely used in molecular diagnostics to make a cDNA copy of an mrna and then PCR reaction amplifies the segment.

Alkaline phosphatase enzyme had its wide use in cloning to remove 5'phosphate from nucleic acids to prevent recircularization of plasmids or DNA vectors used in cloning.

Amylase is a class of starch digesting enzyme which breaks down the complex starch into smaller carbohydrates like maltose. It has no particular role in amplification reaction in molecular biology, but is rather a digestive enzyme.
