Answer #1

Bone Morphogenetic Protein (BMP)4:

It is a highly conserved protein and belongs to transforming growth factor beta super family of proteins. It is involved in bone and cartilage formation. BMP4 plays an important role in the onset of endochondral bone formation. It is also reported to be involved in muscle development, uretic bud development and bone mineralization. BMP4 promotes differentiation of overlying ectodermal tissue. BMP4 shows zygotic expression. It is expressed throughout the late blastula stage and involved in mesoderm ventralization.

Expression of Noggin inhibits BMP signaling in the organizer region. This is essential for the neural induction.

Even though BMP is expressed throughout, it exhibits a signaling gradient. This signaling gradient provides positional information for dorso-ventral patterning of the mesoderm. Low BMP levels specify dorsal tissues while high BMP levels specify ventral tissues.

Head is formed where BMP, Wnt and Nodal signaling are low.

Tail is formed where BMP, Wnt and Nodal signaling are high.

    2. You are studying a (hypothetical) newly-described member of a family of protein kinases (XYK6), which you suspect may function as an oncogene, based upon the observation that specific point mutations (especially Thr489Asp) are sometimes seen in human breast tumors. You’ve queried the TCGA database and found that this mutation, when present, seems to be an “early” mutation, since it is seen as frequently in early-stage lesions as in later-stage tumors. You therefore hypothesize that it may be a driver...

Analyze the data. Investigators discovered a new member of the Bel family. To determine the function of this new Bcl protein, the investigators used cell free extracts. They measured the levels of apoptosis in the cells by measuring the activation of caspase 9 in the cells and determined the effect of addition of the novel Bcl protein to the extracts. The results are shown in Figure A. (the numbers indicate the microgram amount of the novel Bcl protein added to...
