Prokaryotic Promoter:

  • includes two conserved sequences at -35 and -10. This is a feature of the Prokaryotic Promoter. The Alpha factor of the core Bacterial Transcription machinery binds to this sequence and recruits other molecules.
  • recognized by the \sigma factor of RNA polymerase. The \sigma factor is a Prokaryotic transcription factor, involved in Promoter binding

Eukaryotic Promoter:

  • may include a TATA box (not the Pribnow box) upstream of the transcription start site. The TATA box is an Eukaryotic Core-promoter element, with the conserved sequence 5'-TATAAA-3'. The bacterial equivalent is the Pribnow box.
  • May include sequences downstream from the transcription start site. Promoter elements located downstream of the TSS are known as DPE (Downstream Promoter Element). These are known to occur only in Eukaryotes.

Both

  • contains highly conserved DNA sequences. Both Prokaryotic and Eukaryotic promoters are made up of highly conserved sequences. This allows for the regulation of transcription of different genes by the same basal transcription machinery.
  • requires multiple proteins called general transcription factors. General transcription factors are present in both Prokaryotes and Eukaryotes and regulate the rate and timing of transcription.
