Answer #1

Epinephrine receptors are G-protein coupled receptors which is the largest gene family in the human genome. All of them signal through coupling to Gαs G-proteins, adenylyl cyclase , and cAMP-dependent protein kinase A. Binding of the receptor to its ligand induce  a rise in the intracellular level of cAMP, resulting from activation of adenylyl cyclase.

In absence of ligand, the α subunit of Gs protein (G) is bound to GDP. Ligand binding changes its conformation, causing it to bind to the trimeric Gs protein in such a way that GDP is displaced from G and GTP is bound. The G·GTP complex, which dissociates from the G complex, then binds to and activates adenylyl cyclase. The GTP bound to G is subsequently hydrolyzed to GDP leading to the inactivation of adenylyl cyclase. Thus, in the absence of GTP hydrolysis due to binding of GTP gamma S, cAMP would increase due to constant activation of adenylate cyclase by G-proteins.

Epinephrine signaling , like that of insulin, is a multi-step pathway with several amplification steps. Identify each of the steps below as either allosteric activation or enzymatic. Abbreviations: Epi (epinephrine),  β2AR (β2 adrenergic receptor), R2C2 (subunit composition of protein kinase A where R is the regulatory subunit and C is the catalytic subunit), PDE (cyclic AMP phosphodiesterase), PPase (phosphoprotein phosphatase). You are interested in cell size regulation and discover that signaling through a GPCR is important in controlling cell size in rat white blood cells. The G protein downstream of this receptor activates adenylyl cyclase, which ultimately leads to the activation of PKA. You find that cells that lack the GPCR are 15% smaller than normal cells, whereas cells that express a mutant, constitutively activated version of PKA are 15% larger than normal cells. Furthermore, the normal blood cells become...

Drosopholia eye development is an example of cell differentiation directed by signal binding to a receptor tyrosine kinase (RTK). In this pathway, R7 photoreceptor development requires a receptor (SEV, sevenless), which is activated by binding to a plasma membrane bound signal (BOSS, bride-of-sevenless) displayed on the adjacent R8 cell Receptor activation leads to activation of RAS via two proteins, Drk (down- stream of receptor kinases) and Sos (son-of-sevenless). Activated RAS leads to activation of a phosphorylation cascade that activates the...

Question 10-12 10. During the mating process, yeast cells respond to pheromones secreted by other yeast cells. These pheromones bind GPCRs on the surface of the responding cell and lead to the activation of proteins inside the cell. When a wild-type yeast cell senses the pheromone, its physiology changes in preparation for mating: the cell stops growing until it finds a mating partner. If yeast cells do not undergo the appropriate response after sensing a pheromone, they are considered sterile....
