  1. Why do normal T cells, which recognize foreign peptide antigens bound to self MHC molecules, react strongly against the allogeneic MHC molecules of a graft?

Allogeneic MHC molecules with any bound peptide are probably going to resemble self MHC plus a foreign peptide, hence normal T cells might cross-react with the allogeneic molecules. There are thousands of copies of allogeneic MHC molecules on every graft cell showing different donor peptides. Several of these peptide-MHC complexes may be recognized by one of the many thousands of clones of a graft recipient's T cells. Individuals develop tolerance to self-antigens but do not tolerate the foreign antigens of a graft.
