Answer: The enzymes which is generally used to map the region of open chromatin is called as Nuclease specially DNase1 and MNase (micrococcal nuclease) .

Explanation: Firstly i like to clear what is chromatin so---

Chromatin = DNA+ Protein means chromatin is the structure where DNA is wrapped around a protein. The protein present here is called as histone protein.

DNase enzyme can act on the DNA strand vigrously irrespective to sequence specificity and can cleave off the DNA strand. When the DNA sequence is bound with protein then in those case there enzymes can not attack the DNA so nuclase DNase 1 is used to cleave the phosphodiester bond between hydroxyl and phosphate group. DNase generally work on SsDNA, DsDNA, as well as in  chromatin.

The other type of nuclease is called as  micrococcal nuclease(MNase) which is basically acting on single stranded nucleic acid.
