Problem 5. True/False.
(Is the statement true or false? Explain why.).

1. Both Northern blot and in situ hybridization techniques start with extracting and
purifying all transcripts from an organism, an organ, or a group of cells.

2. Both Northern and Western blot techniques require preparation of a labeled
DNA probe that is complementary to the transcript of a gene that you study

Answer #1

Q1)Answer:-False statement because northern blotting techniques start with extracting and purifying transcript from an organism ,an organ , or a group of cells and then electrophoresis is done to separate RNA molecules based on their size while in insitu hybridization these steps are not done as in this technique specific mRNA inside cell or tissue is localised by using DNA probe within the cell or tissue.

Q2)Answer:- False statement in Northern blotting technique only required preparation of a labelled DNA probe that is complementary to the transcript of a gene that we study but in Western blotting primary antibodies are used to the nitrocellulose or PVDF membrane to bind with protein present in the membrane and after the binding of primary antibodies and protein of desired interest ,they are incubated in the secondary antibodies.
