When you are attempting to focus the microscope on a specimen, you always start with the stage all the way [stage] and the [objective] objective in position above the specimen.

The correct option is (E) down....low power
So when a specimen has to be focused in a microscope, it is first kekpt on the stage and the stage is kept at its very low position...Also the objective lens is kept at its low power which is 4x, just to have a rough idea about where the specimen is present on the slide. Then, afterwards the stage is pulled upwards keeping your eyes in the low power lens. When you see a slight reflection of the specimen..then the fine knob comes in help..you roll the fine knob as much it is necessary to get the clear picture of the specimen. Then you start switching to high power lens to get a more clear picture.
