11. What effect does an insertion mutation have on the mRNA reading frame, give an example?
Insertion means addition of a new nucleotide.
If one or two bases are added, then the whole amino acid sequence is changed. If 3 bases are added then one new amino acid is introduced and of the amino acids remain same.
Example,
Wild type mRNA sequence,
5'- AUG CGU AGC GAA UAU UUA -3'
Wild type amino acid sequence will be ,
N- met arg ser glu tyr leu -C
Case 1 - insertion of A at postion 10
5'- AUG CGU AGC AGA AUA UUU A -3'
Mutated protein, N- met arg ser arg ile phe -C
Case 2 - insertion of AG at position 10 and 11 respectively
5'- AUG CGU AGC AGG AAU AUU UA -3'
Mutated protein, N- met arg ser arg asn ile -C
Case 3 - insertion of AGU at position 10, 11 and 12 respectively
5'- AUG CGU AGC AGU GAA UAU UUA -3'
Mutated protein, N-met arg ser ser glu tyr leu -C
In this last case, serine is inserted, rest will remain same.
Please rate.
Mutations are given in bold.
11. What effect does an insertion mutation have on the mRNA reading frame, give an example?
11. Match each type of mutation with the corresponding description. (4 points) Missense An insertion or deletion of nucleotides that are not in multiples of 3. Nonsense A mutation that does not alter the protein sequence. Silent A mutation that confers an amino acid substitution. Frameshift A mutation that confers a premature stop codon.
Take Test: HW 4 QUESTION 16 What type of mutation leads to frame shift of dna? O A. Deletion mutation B. Insertion mutation C. Point mutation OD. None
What are the potential consequences of inaccurate mRNA splicing? a. A single base error at a splice junction will cause a large deletion. b. A single base error at a splice junction will cause a large inversion. c. A single base error at a splice junction will cause a large insertion. d. A single base error at a splice junction will change the reading frame and result in mRNA mistranslation.
What is the primary reason that only a single reading frame is used during translation of a given monocistronic mRNA.
Is ncRNA linked to mRNA by covalent bonds, base complementary or both? What effect does this have on the transcript?
Original DNA & mRNA: DNA template: 3' - GGGTACGGCTCTAAA(300b)TACCCGATCGTA - 5' mRNA: 5' - CCCAUGCCGAGAUUU(300b)AUGGGCUAGCAU - 3' What effect would the following DNA mutation have on the polypeptide synthesized from the gene? DNA template: 3' - GGGTTCGGCTCTAAA(300b)TACCCGATCGTA - 5' mRNA: 5' - CCCAAGCCGAGAUUU(300b)AUGGGCUAGCAU - 3' There will be no effect because the codon still calls for the same amino acid. The polypeptide's synthesis will stop too soon because a stop codon has been introduced too early in the sequence. A...

Suppose that there is a frameshift mutation between the 10^th and 11^th base (counting from the left: a Guanine is added) in the minus strand of DNA, finish the table giving the resulting peptide. Circle the changes in the resulting peptide caused by the mutation. What is the importance of these changes. (Assume the reading frame starts with the first codon)
2. If a mutation occurs and alters the sequence CGATATGA to CGATATCA what will be the final effect - compared to the protein that was supposed to be made, how would the actual protein that is synthesized (made) look -shorter, longer or same? 3. Give the sequence of the protein that would result from such a mutation (mutation in question 4. What is the name given to such a mutation? Be specific (answer is not point mutation, deletion, insertion, substitution...
