  • What might be wrong with the 1X TAE for an agarose gel to max out the amperage (I) and result in a lower voltage (v). (pH 8.3)

Th ions which are provided by TAE buffer were not sufficient to carry out the electricity for electrolysis and the components are not functioning properly like EDTA to chelate divalent ions as a result the ions are not enough in the Gel for proper electrolysis process and thus amperage and voltage difference decreases resulting in failure of separation of DNA.

So , here 2× or more TAE must be used.
