Most likely sex of the individual from which those cells have been taken is female.

Males have only one X chromosome, whereas females have two X chromosomes. To keep the gene expression levels same in both the organisms, system evolved to silence the extra X chromosome in females through X-inactivation. This extra X-chromosome becomes heterochromatin and appears as a Barr body. So only females should have one Barr body. Some females with 47, XXX will have two Barr bodies.

Only exception for this is the Klinefelter syndrome (47, XXY), where the person is male because of Y chromosome, but Barr body will be there to silence the extra X chromosome.
