Melting temperature of DNA primer depending on the DNA sequence. Those DNA strand contain more G+C content have high melting temperature(Tm).

Melting temperature can be calculated the following formula :

     Tm= (wA+xT) * 2 + (yG+zC) * 4

     where w,x,y,z are the number of the bases A,T,G,C in the sequence

here more G and C content sequences is options C sequence, contain G+C =11 .so this sequence have high melting temperature.
