A serial diution as performed using 100 microliters of the phage and 900 microliters of LB. You should reach a maximum dilution of 10^-6. Using the exact protocol as you did in lab, what would the titer of the virus stock be if you observed 73 plaques on the plate with the greatest dilution?

Answer #1

Plaque-forming units, PFUs for viral counts :

Plaque-forming units, PFU/ml = No. of Plaques / dXV                                           d = dilution

V = volume of diuted virus added to the plate.

Plaque-forming units, PFU/ml =    73 / 10-6

              Titer of the virus stock =    73 \times 10 6 PFU/ml
