For the increase in the expression of the VEGF which is vascular epithelial growth factor and helpful in the angiogenesis in the cancer cells in the higher amount depends on the expression of the HIF1a that is the hypoxia indicuble factor 1 alpha that is helpful in the expression dynamics of the VEGF. It has been seen that the binding of the prolin in the hypoxia condition to HIF1a can lead to the rapid degradation of the HIF1a and ultimately reduces down the expression for VEGF as well.

Thus it is important that the affinity of the proline towards the HIF1a has to change thus, the conversion of the proline to alanine will change the overall affinity of the ligand towards the macromolecule and thus lead to the increase in the HIF1a expression as well that will lead to the increase in the VEGF gene expression too.
