You perform interrupted conjugation using an a+b+c+d+e+f+g+h+ Hfr strain and an F strain that is abcdefgh. You observe the following genes transferred together in order from last to first:

                                 h+b+g+d+

e+f+d+g+

f+e+a+c+

What is the map order of the genes?

Answer #1

ANSWER

Let us label the fragments as 1, 2 and 3.

1 →   h+ b+ g+ d+

2 →   e+ f+ d+ g+

3 → f+ e+ a+ c+

Now let us try to overlap the fragments. If you observe, you’ll notice that fragment 2 does not overlap with the other two fragments. Therefore, we need to invert fragment 2.

htbt gt dt gt d+ ft et ft et at ct

From the above comparison, we can conclude that the map order of the genes is:

h+ b+ g+ d+ f+ e+ a+ c+

The giant sandworms of the planet Arrakis harbor a gut bacterium that undergoes conjugation. However, the conjugation lasts for 200 minutes. You perform an interrupted mating experiments and find that following markers are transformed at the respective times: ple 25 minutes, bar 75 minutes, gap 100 minutes, zap 150 minutes and chr 200 minutes. Draw a genetic map of the bacterium assuming a linear transfer rate of DNA. You also find that the plemarker is transferred at a higher rate...

An Hfr strain is used to map four genes in an interrupted mating experiment. The cross is  Hfr/a+b+c+ rifs ×F−a−b−c− rifr . (No map order is implied in the listing of the alleles; rifr is  resistance to the antibiotic rifampicin.) The a+gene is required for  the biosynthesis of  nutrient A, the b+ gene for nutrient B, c+ for nutrient C. The cross is initiated at time = 0, and  at  various times, the mating mixture is plated on medium. Each plate contains minimal  medium (MM) plus rifampicin....

Three Hfr strains of E. coli, each with genotype StrS, lacZ+, A, B, C, D are mated to an F- strain with the genotype StrR, lacZ-, a, b, c, d. Samples of conjugating strains are collected and disrupted every minute for one hour using a blender. Cells are plated under conditions that allow the lacZ+, A, B, C, and D alleles to be identified.
