Intense exercising cause anaerobic conditions in muscles. In the absence of Oxygen- the terminal acceptor of electron ATP's cannot be generated through electron transport chain. hence muscles switch to lactate fermentation to keep glycolysis running and to generate ATP by substrate-level phosphorylation.

NAD+ is essential for glycolysis, in the absence of oxygen, lactate fermentation sets in. The NADH formed is oxidized back to form NAD+ to keep the glycolysis under anaerobic conditions.
