I believe the correct answer to be:

1) Option B) seperate the two DNA straas of the template.

As denaturation which occurs at high temperatures seperate the 2 DNA strands that were previously held together by hydrogen bonds.

2) Option B) Regulatory sequences.

As they play a role in transiption which is not needed by cloning vector but are needed by expression vector as they need to express the gene present in them.
