Disc diffusion test is done to check the antibiotic sensitivity to the bacteria. In test, bacterial agar plat is used. On agar plate , antibiotics are applied by swabs and then incubated at room temperature overnight. If the plates, seeded with test strains are left longer than optimum period of time. Than there may be multiplication of inoculum nest by the disc this will cause reduction in zone diameter. This may report test strain as resistant strain.
