  2. For Microbiology lab class. The Schaeffer-Fulton stain is used for endospore staining. I do not understand the question and what it is asking for. I do know that the primary stain is what stains the endospore to malachite green.

In the question, it has been asked, what is the dormancy structure present in bacteria that is observed using schaeffer- Fulton method and that can be used to differentiate populations of bacteria?

The staining method is used for observing Endospores produced by certain bacterial species. Endospores are structures that a bacteria produces during harsh conditions, therefore providing dormancy to that particular bacteria. Not all bacterial strains can produce endospores, therefore this staining method can be used to distinguish between bacterial strains based on their ability to produce endospores.

In this method Primary stain Malachite Green along with heat is used to stain the endospores. This gives the spores dark green color and the vegetal cells are observed pink.
