Cation exchange column is having –vely charged matrix which exchanges or attracts +vely charged molecules and the molecules above their pI are –vely charged and below their pI are +vely charged, in the question we need +vely charged molecules hence the sutable pH is 3.9 where both molecules exist in +ve charge

Because the proteins are in the range of 2000-7000-20000 and 90000, 90k protein comes in void volume and the first to get elute followed by 20k, 7k and the last one 2k

Because spf. Activity = number of units/ protein in mg
