1) basic dyes are positively charged and the bacterial cell wall is negatively charged so basic dyes can stain the bacterial cell walls more efficiently because positive and negative charges attract each other.

2) resolution of is the minimum distance (r) between 2 points such that the 2 points can be distinguished as 2 separate points,

r= 1.22/2nsin, where = wavelength of light, 2nsin= numerical aperture of the lens

n is the refractive index

oil has greater refractive index than air ( n of air=1) if we are using oil as the medium, refractive index of medium increases and r is inversely proportional to n so as n increases r decreases,

so resolving of the microscope increases so we can clearly see the the bacteria.
