Explanation:

From the 1ml of 2x10⁸ cells/ml add in to 9 ml of normal saline (now dilution is 10 times 2x10⁷), = 1ml+9ml (10 times dilution)

From above diluted sample take 1ml and add in to fresh 9ml normal saline (now dilution is 100 times 2x10⁶) = 1ml+9ml (100 times dilution)

From above diluted sample take 1ml and add in to fresh 9ml normal saline (now dilution is 1000 times 2x10⁵). = 1ml+9ml (1000 times dilution)

From above diluted sample take 1ml and add in to fresh 199ml normal saline (now dilution 2x10⁵/200 or 2x10⁵/2x10² = 1x10³).
