Answer: The culture you serially dilute up to 2x105 and then dilute it 200 time you to obtain 1x103 cells per ml.

Explanation:

  1. From the 1ml of 2x108 cells/ml add in to 9 ml of normal saline (now dilution is 10 times 2x107), = 1ml+9ml (10 times dilution)
  1. From above diluted sample take 1ml and add in to fresh 9ml normal saline (now dilution is 100 times 2x106) = 1ml+9ml (100 times dilution)
  1. From above diluted sample take 1ml and add in to fresh 9ml normal saline (now dilution is 1000 times 2x105). = 1ml+9ml (1000 times dilution)
  1. From above diluted sample take 1ml and add in to fresh 199ml normal saline (now dilution 2x105/200 or 2x105/2x102 = 1x103).
