Some mutations occur in the promoter of Msh2 and decrease its expression relative to wild-type cells leading to cancer. Would you confirm this pattern using DNA sequence or RNA sequence and why?

DNA sequence.

The promoter is present upstream of the transcription start site. So it is not transcribed ding the transcription.

If we want to confirm that there is a mutation in the promoter then we need to perform DNA sequencing.
