High throughout DNA sequencing by synthesis is also known as Next-generation sequencing.

It involves following steps.

1. DNA cleavage to produce small fragments.

2. Primer extension in the presence of labelled and blocked nucleotides

3. Separation by electrophoresis and read out of labels.

So, it uses ddNTPs.
