The RNA polymerase II builds the RNA that codes for proteins. Cellular differentiation is determined by the expression of certain areas of the genes. All DNA has the coding for that cell which potentially differentiate cells. The regulation of portion of the DNA controls the cell's function.

Consider a cell from liver. Then the part of DNA that encodes for proteins from liver cells will be transcribed and translated. The specific coding for this lies in the regulation RNA polymerase II. Once it transcribes the correct mRNA sequence and proper protein.
