The traditional method for calculating protein concentration of an unknown sample is to use a standard curve that is generated from known protein standards.   A linear regression for the set of standards as they assume the overall relationship between the standards is best described by a linear relationship (R2=0.9791), which is rarely seen. The red line in the Linear Plot figure shows the linear plot and the equation for this line is y=1.0736x + 0.0677. Solving for x for an absorbance will give the protein concentration

a) for 10 ul, the absorbance is 0.2, so the amount of protein concentration

x= (Y-0.0677)/1.0736=( 0.2-0.0677)/1.0736=0.12 ug

for 20 ul the protein amount is 0.29 ug

for 40 ul the protein amount is 0.63 ug

B) since the dilution was 1:100

the total amount of protein in each reaction is

0.12x100=12ug

0.29x100=29ug

0.63x100=63ug.
