Answer #1

Figure 2:

Question - What is the cellular localulocali of GR protein when treated with dexamethasone and arsenic compared to control cells?

Result - In control (untreated) cells, glucocorticoid receptor (GR) seems to be localized predominantly in the cytoplasm of the cells whereas when treated with dexamethasone and arsenic, GR is found to be present only in nucleus.

(When stained with both GFP and PI, which is a nuclear stain, the dexamethasone and arsenic images show yellow nucleus instead of red, which means the green of GFP and red of PI had combined to give yellow. Since green is not seen anywhere else, GR is present solely in nucleus in dexamethasone and arsenic treated cells)

    2) Given that E6 inactivates p53-dependent pathways, cervical cancer cells are particularly resistant to traditional treatments such as chemotherapy, the search is ongoing for new alternative therapeutics to be used in conjunction with current chemotherapeutic drugs. One such candidate is the plant flavonoid Luteolin. In 2014, Ham et al evaluate the effectiveness of Luteolin on sensitizing cervical cancer cells to apoptosis. a) In Figure 1, panel A, the researchers looked for an effect on E6 levels post Luteolin treatment. Total question is worth 10 marks A1. Answer ALL parts: In insulin-responsive cells the insulin receptor substrate 1 (IRS-1) protein is believed to be a major component of signal transduction. Several experiments were performed to investigate the relationship between IRS-1 and three adaptor proteins (Grb2, Shc, and p85) with respect to their role in insulin signaling. The descriptions of experiments are provided in the legends to the figures. Use Figures 1, 2 and 3 to provide the most appropriate answer....
