Actually simple staining procedure incluid various steps.

one of them is fixation of cell. in this step we just make the sample immovable in order to study them properly, because purpose of simple staining is to study their structural part not about their movement.

So inorder to fix them (immobilize), here different grades of alcohol were used, which dehydrate the sample only to fix them at a particular stage. As there is dehydration of cell occurs, cell lost all the water content which ultimately cut all the connection of that cell with nature and cell(sample) become die.

Then there is application of colour (staining step). in case of simple staining mainly basic stains are used like methylene blue etc...

Basic stains are +vely charged so they tends to bonded with -vely charged alcohol treated sample (i.e. stains only attached to -vely charged part of bateria), which faciliates us to show the structural part of bacteria whereever the stains get attached.

SO THE SIMPLE STAIN WILL DYE BACTERIA ONLY BECAUSE OF OPPOSITE CHARGE INTERACTION WHICH FACILIATES STRONG INTERACTION BETWEEN STAIN AND BACTERIA.

THE BACTERIA GET DIE ONLY BECAUSE OF DEHYDRATION STEP &
