Amylase break down starch into maltose. So the substrate of the enzyme is starch. More specifically it is two compounds amylose and amylopectin which is found in starch.

Active site is the region of enzyme where substrate bind and undergo chemical reaction.

Since the body temperature is 37 0C, the maximum optimum temperature for the amylase activity is 37 0C. Optimum temperature range is 35 to 37 0C

Optimum pH range of amylase is 6.7–7. The normal pH range for saliva is 6.2 to 7.6. Amylase will function in this range.
