There are proton pumps located on the membrane of synaptic vesicles that generate gradient required for the transport of neurotransmitter. Most commonly found proton pump is V-ATPase that uses energy to transport protons in vesicle lowering the pH of vesicle. The antiporter uses the energy stored in the gradient of protons to transport neurotransmitter inside the vesicles. They transport protons out of the vesicle while moving neurotransmitter in the vesicle.

So option B (oh in the vesicle is lower than cytoplasm) is correct
