We can take that 2 samples of that plant and grow it in different light duration and observe the growth of both the samples.
In which more growth is occuring, we can detect according to its growth that whether it is growing better in more light or less light and accordingly we can decide whether it is long day or short day.
To see whether harmone is involved in root formation, we will add inhibitor of that harmone and see if root formation is normal or there is some changes. If root formation do not occur or deviates from normal it means harmone indeed was involve in root formation.
We can also detect this, but artificially providing more harmone (same harmone) to plant and see it's effect on growth formation.
