Why is de-ionized (ion-exchange) water not suitable as a solvent for use in UV absorbance measurements?
Answer
De-ionized water is described as the water that lacks ions like sodium, calcium etc. The ionic impurity is removed in Deionised water hence it has low conductivity.
In the case of contamination of plasticizers or organic impurity, the water quality will be degraded although the conductivity will not rise. the total carbon content in deionized water will be high. Measurement of water conductivity for quality check of deionised will be meaningless. The pure water will have low conductivity and TOC hence MilliQ is recommended as a solvent for UV absorbance measurement. It also has low contamination than deionized water and will not show any UV absorbance peak.
As high UV absorbance is observed in deionized water due to its conductivity, it is not suitable as a solvent for use in UV absorbance measurement.

Why is it a bad idea to use polystyrene cuvettes when making UV absorbance measurements in the 200-300 nm range? Choose one. a. Polystyrene will be attached by acid. b. Polystyrene absorbs a lot of light in this range. c. Polystyrene might react with some species in the sample. d. Polystyrene fluoresces in this range. e. Polystyrene will be attacked by some organic solvents.

a) Why do you use a minimum of hot water solvent in carrying out a recrystallization of a particular compound? b) Why is it desirable to use suction filtration rather than gravimetric filtration in recovering the purified crystals? c) Why should the suction be released before the solid is washed with fresh solvent?
There are few questions I wanna ask about the Ion-exchange chromatography. 1. Why we measure the absorbance of the protein fraction at λ280 after purification by IEX? 2. After IEX, we are required to carry out BCA assay and L-DOPA assay to determine the protein's concentration and activity but why the instruction said I had to choose the fraction with highest absorbance value? 3. Can you think of one application of IEX to our daily life?

Why can we not use water as a solvent for the reaction of cyclopentadiene and maleic anhydride? Is it possible to convert between the two products without using the corresponding precursor ( ethyl acetate) ? How?
14. (3 pts.) The ionized benzoic acid dissolves better in water than it does in diethyl ether, which is how and why it is extracted into the water phase. Give the most important IMFs that are responsible for solubilizing the benzoic acid anion in the water. Give also the most important IMFs that are responsible for solubilizing the sodium cation in the water. The kinds of strong IMF that could be present are ion-dipole, hydrogen bonding (ignore dipole-dipole and induced...
For TLC plate separation, why do we use Rf values instead of distance measurements (e.g. the spot travelled 5 cm from the starting line)? What would happen if we compared solvent systems using distances rather than Rf values?
