Question number 1 : Body tube of the microsope is the long tube that holds the eye piece/ocular lense and connects the objective lense. This body tube can be raised or lowered by the use of fine and coarse adjustments. But, while looking through the ocular lense it should not be changed (should not be lowered or raised). Once focussed it should not be changed as it increases the risk of hitting the lense and damaging it and it also has the risk of objective lens getting in contact with the slides being observed.

Question number 2 : A diaphragm on a microscope is the piece that enables the user to adjust the amount of light that is focused under the specimen being observed and also create contrast for better viewing transparent specimens.
