No, it would not be appropriate to determine the concentration of cells at 500 nm using a curve drawn with Ab 600 values. This is because the absorptive properties of cells would differ between 500 nm and 600 nm and may not be directly correlated. One reason for this is that the cellular constituents that absorb at 600 nm may not absorb at 500 nm and the concentration of these substances might vary between cells at high density and low density.
