The characteristic difference between acid fast bacteria and other organisms is the presence of a thick, waxy wall (made up of mycolic acid) that makes penetration by stains very difficult. Once the stain has penetrated, however it cannot be readily removed even with the vigorous use of acid alcohol as the decolorizing agent, because of this property these organisms are called acid fast, while all other microorganisms which can be easily decolorized by acid alcohol are called as non-acid fast.

ACID FAST BACTERIA WILL BE RED IN COLOUR

NON-ACID FAST BACTERIA WILL BE BLUE IN COLOUR
